Mammals experience some degree of hypoxia during their lifetime. In response to hypoxic challenge, mammalian cells orchestrate specific responses at transcriptional and posttranslational level which lead changes in the purine metabolites order cope with threatening conditions. The aim this study was evaluate enzymes involved metabolism human muscle Muscle culture were exposed enzymatic activity inosine monophosphate dehydrogenase (IMPDH), xanthine oxidase (XO), nucleoside phosphorylase (PNP) hypoxanthine guanine phosphoribosyl transferase (HGPRT) as well transcript expression quantified under normoxic Purine metabolite (hypoxanthine (HX), (X), uric acid (UA), (IMP), inosine, nicotinamide adenine dinucleotide (NAD+), adenosine, adenosine (AMP), diphosphate (ADP), triphosphate (ATP), guanosine (GDP) (GTP)) concentrations also quantified. Significant reduction IMPDH HX IMP (p < 0.05) observed after hypoxia, suggesting a decrease de novo synthesis purines. After global transcripts observed, metabolic machinery new steady states that balance ATP demand supply pathways.
